Hi !
On page 137 Bascom Avr PDF help...
But, in few steps:
First connect Arduino Atmega2560 with usb to your PC, than in Bascom menu OPTIONS chose Programmer, than chose: ARDUINO STK500V2,
proper COM port (in device manager can find which com is used for Arduino...) and for baud 115200...
work like normal programmer...

What is the baudrate you are using?
Are you using a cable on the ISP connector or the bootloader?

What is the baudrate you are using? 9600 and 115200
Are you using a cable on the ISP connector or the bootloader?
Use USB cable

Have you loaded the driver for the M2560 onto your computer?

As I said in my tutorial
FIRST the 2560 arduino must be work with arduino compiler, here : https://www.arduino.cc/en/Main/Software then you must see the port used in the parameters of your computer.
if the small blink program gave in the sample arduino doesn't work, you can reload the driver from Arduino IDE
THEN and only if the steps above are working you can work with Bascom IDE
Did you follow it ?
Are you use to use the good serial port ? (in the picture the Arduino is not recognized.) then If you use the autoflash you must wait until bascom see your Arduino board
